The problem: NAD+ levels fall roughly 50% between age 20 and 60 in most human tissues, measured in both blood and muscle biopsies. This is not a marginal decline. The consequences are proportional. NMN (nicotinamide mononucleotide) is a direct precursor to NAD+. It is absorbed primarily via the Slc12a8 intestinal transporter and converted to NAD+ in tissues through the action of NMNAT enzymes. The conversion is efficient and well-characterized.
The landmark Yoshino 2021 trial (published in Science) demonstrated that 250mg/day of NMN improved skeletal muscle insulin signaling in postmenopausal women with prediabetes, providing the first robust human evidence of clinically meaningful tissue-level effects. Multiple subsequent trials have confirmed measurable NAD+ elevation. The Huang 2022 GeroScience trial found dose-dependent improvements in gait speed and grip strength at 600mg/day — exactly the functional markers that predict longevity outcomes.
There is, however, a critical purchasing problem: ConsumerLab independent testing found actual NMN content ranging from 12% to 107% of label claims across commercial products. That is a 9-fold spread. A nominally 500mg product could be delivering 60mg of actual NMN. Purity verification is not a nice-to-have — it is the most important variable in the purchase decision, ahead of dose, delivery form, or price per gram.
Not all third-party testing is equivalent. The term appears on many NMN labels but can mean anything from a rigorous pharmaceutical-grade audit to a single-batch identity test performed once at launch. Here is what meaningful verification actually involves:
Actual milligrams of NMN vs. stated label amount, verified by HPLC chromatography. This is the critical test. Anything less than 90% of label is a failure.

Most human trials administered NMN in the morning. There is mechanistic support for this timing: NAD+ participates in circadian clock regulation through SIRT1-mediated BMAL1 and CLOCK deacetylation, and some practitioners report sleep disruption at evening doses. Morning with or without food is the most evidence-consistent protocol. Some practitioners pair NMN with a small amount of fat for potential absorption benefit in capsule formulations, though this is not established in direct human comparative trials. These are functional outcomes that correlate with healthspan in population data. No human trial has yet measured lifespan — that is an animal study extrapolation and a category of evidence we should not overweight. What we can say with reasonable confidence: NAD+ decline is mechanistically linked to multiple aging hallmarks, NMN reliably reverses that decline in a dose-dependent fashion, and the downstream functional effects in older adults are measurable and consistent across multiple independent research groups. It is among the most evidence-backed longevity supplements currently available, with the important caveat that “evidence-backed” in this category still means early-stage human trials, not decades of outcomes data.
