Collagen peptide supplements are one of the best-selling categories in the longevity and beauty-supplement space, and individual placebo-controlled trials genuinely do report skin benefits. The complication is a 2025 meta-analysis that did something most supplement literature doesn't: split the evidence base by who paid for each trial, and found the picture changes substantially once you do.

The Individual Trial Evidence — Proksch 2014

The most-cited direct evidence for collagen peptides and skin comes from two related 2014 trials by Ehrhardt Proksch and colleagues, published in Skin Pharmacology and Physiology.

Key Finding: In the first trial, 69 women aged 35-55 were randomized to 2.5g or 5.0g of collagen hydrolysate or placebo once daily for 8 weeks; researchers measured skin elasticity, moisture, transepidermal water loss, and roughness objectively. In a second, related trial, 114 women aged 45-65 were randomized to 2.5g of a specific bioactive collagen peptide (marketed as VERISOL) or placebo daily for 8 weeks, with eye-area wrinkle depth as the primary outcome. Both trials reported statistically significant improvements favoring collagen peptide supplementation.

The 2025 Meta-Analysis — Myung & Park, American Journal of Medicine

Individual trials like Proksch 2014 are exactly the kind of study that gets rolled up into meta-analyses to estimate an overall effect size. When Seung-Kwon Myung and Yunseo Park did this across the full published literature, the initial pooled result looked like more good news for collagen.

Key Finding: Myung and Park (2025, American Journal of Medicine) pooled 23 randomized controlled trials covering 1,474 participants. The initial pooled analysis showed collagen supplementation linked to measurable improvements in skin hydration, elasticity, and wrinkle reduction -- consistent with Proksch 2014 and similar individual trials.
Honest limitation — and the actual headline finding: The researchers then found that 21 of the 23 trials in that evidence base had some form of industry funding or sponsorship involvement. When they split the analysis by funding source, independently funded trials showed no significant benefit for hydration, elasticity, or wrinkles in any outcome category, while industry-funded trials consistently reported significant positive effects. The authors' conclusion: there is currently no clinical evidence to support the use of collagen supplements to prevent or treat skin aging, once funding-related bias is accounted for.

This is not the same as saying collagen peptides "don't work" in some absolute sense -- with only 2 of 23 trials independently funded, the independent evidence base is too thin to be conclusive in either direction. What it does mean is that the existing published literature, taken as a whole, is more consistent with funding-related bias driving the reported effect sizes than with a robust, funding-independent biological signal.

What About Joint Health?

Oral collagen peptides are also widely marketed for joint comfort and cartilage support, distinct from the skin-aging claims covered above. That evidence base is separate from the Myung & Park skin meta-analysis and generally smaller, with similar concerns about industry sponsorship common across the sports-nutrition supplement literature more broadly. This guide focuses on the skin-aging evidence specifically, since that's where the most direct funding-bias analysis exists; treat joint-health claims with the same baseline skepticism about sponsorship until an equivalent independently-funded meta-analysis exists for that outcome.
